What is Ipamorelin
Ipamorelin (Aib-His-D-2-Nal-D-Phe-Lys-NH₂) is a pentapeptide growth hormone secretagogue developed in the late 1990s. It acts on the ghrelin receptor (GHS-R1a) — different from the GHRH receptor that CJC-1295 acts on.
Its defining characteristic is selectivity: a classic study (Raun, 1998) showed that Ipamorelin releases GH at levels comparable to GHRP-6, but without the cortisol, prolactin, or ACTH release characteristic of older secretagogues. It does not increase cortisol or ACTH even at 200 times the effective GH-releasing dose — which is why it has become the preferred GHRP in combined studies.
How it Works
Ipamorelin binds to the ghrelin receptor (GHSR1a) on pituitary cells and triggers calcium-dependent GH release, which amplifies the current GHRH signal — the "releasing" side of the pulse. In combination with CJC-1295 (the "production" side), the two produce a synergistic effect.
